BAM-15

BAM15 is a mitochondrial protonophore uncoupler, a compound that disrupts the mitochondrial proton gradient to increase energy expenditure without producing ATP

This “uncoupling” of oxidative phosphorylation boosts mitochondrial respiration, leading to potential therapeutic benefits. It has shown promise in:

Obesity: Increases fat burning and reduces body fat mass without affecting food intake or body temperature, potentially aiding weight loss

Diabetes: Improves glycemic control and insulin sensitivity in preclinical studies

Kidney Injury: Protects against acute renal ischemic-reperfusion injury and sepsis-related kidney damage

Cancer: Inhibits proliferation of acute myeloid leukemia (AML) cells by increasing reactive oxygen species (ROS) production

Cardiovascular Disease: Low doses may protect the heart by activating STAT3, though high doses could reduce ATP production

Sepsis: Reduced inflammation and organ damage

Unlike older uncouplers like DNP, BAM15 is less toxic, doesn’t depolarize the plasma membrane, and has a short half-life, reducing buildup risks
